چکیده
Defensin is a gene family with antimicrobial peptides and thought to be effector molecules in innate immunity. Defensins are cationic, cystein-rich antimicrobial peptide components of the mammalian innate system1. They are members of a supergene family consisting of αand β‚ subtypes, located in a cluster at chromosome 82. Human -Defensin-2 Expression by Keratinocytes is Induced by Co- Culture with Trycophyton rubrum Through Toll-Like Receptors 2 and 4
To determine a role of human -Defensin-2 (HBD-2) in defense against cutaneous dermatophyte infection, we investigated the induction of HBD-2 mRNA expression by a nontumorigenic human keratinocyte-derived cell line, HaCaT cells, after co-culture with microconidia collected from the colonies of Trichophyton rubrum. Co-culture of T. rubrum or Candida albicans with HaCaT cells significantly induced...

The human beta-defensin 3 (hBD-3) is an inducible epithelial peptide antibiotic that has potent antistaphylococcal activity. Infection of skin epithelial cells with viable Staphylococcus aureus, a common skin pathogen, induces increased gene expression of hBD-3 and other antimicrobial peptides. The aim of this study was to identify signaling pathways and nuclear responses that contribute to the...
متن کاملMouse beta-defensin-14, an antimicrobial ortholog of human beta-defensin-3.
Searching the database for mouse homologs of the antimicrobial peptide human beta-defensin-3 (hBD-3) revealed highest identity (69%) to mouse beta-defensin-14 (mBD-14). Recombinant mBD-14 exhibited broad-spectrum, nanomolar microbicidal activity. Treatment of keratinocytes with gamma interferon or transforming growth factor alpha increased mBD-14 gene expression. These data suggest that mBD-14 ...
متن کاملSphingosine Kinase-1 Is Required for Toll Mediated β-Defensin 2 Induction in Human Oral Keratinocytes
BACKGROUND Host defense against invading pathogens is triggered by various receptors including toll-like receptors (TLRs). Activation of TLRs is a pivotal step in the initiation of innate, inflammatory, and antimicrobial defense mechanisms. Human beta-defensin 2 (HBD-2) is a cationic antimicrobial peptide secreted upon gram-negative bacterial perturbation in many cells. Stimulation of various T...
متن کاملThe Antimicrobial Peptide Human Beta-Defensin-3 Is Induced by Platelet-Released Growth Factors in Primary Keratinocytes
Platelet-released growth factors (PRGF) and its related clinically used formulations (e.g., Vivostat Platelet-Rich Fibrin (PRF®)) contain a variety of chemokines, cytokines, and growth factors and are therefore used to support healing of chronic, hard-to-heal, or infected wounds. Human beta-defensin-3 (hBD-3) is an antimicrobial peptide inducibly expressed in human keratinocytes especially upon...
